Aseptic- allow handling of materials without the introduction of contaminants for growth of pure cultures (Koch’s Postulates)
- minimize contact with room air and non-sterile surfaces. Keep petri dishes & test tubes closed. Sterilize forcepts, loops, wires, etc...
Agar- solidifying agent (not nutrient). Melts at 100o and solidifies at 45oC.
